The obvious answers here are, for an american master, Eric Roberts, for an american companion, Daphne Ashbrook, and for an american doctor, Paul McGann doing an accent.
That silliness aside, I don't have a tonne of thoughts on companions - in some ways, that's the easiest one to fill! A few doctor thoughts: I'd actually really love to see what Steve Buscemi did with it. D'arcy Carden could have a lot of fun with it. I think Joseph Gordon Levitt might be a slightly boring/safe choice, but he'd probably be good. Jamie Lee Curtis. *Definitely* Janelle Monae. Stephanie Beatriz would also play a really interesting doctor (or maybe master.)
Peter Dinklage would probably be also be good as the doctor, but my God, I see him as an absolutely perfect master. Viola Davis as the master - enough said. Since I mentioned D'arcy Carden earlier, based purely on his performance in the good place, Ted Dansen. Michelle Yeoh (not really american, but has lived and worked there for a long time). If you wanted to go back to a very ainley/delgado esque master, Michael Shannon. And I think that Giancarlo Esposito is almost cheating as a master answer, because he's known at this point for being very good at playing calculating villains.
Sidebar: she's Australian, but Toni Collette as either. As both, possibly.

Peter Dinklage would be my cast. He is an amazing actor and while he is American, he is from Irish (and German) descend, so he has some connection to Brittain at least. He is great at playing quick witted character and his role on Game of Throne shows that he has the range needed for playing the role.
Companion is harder. Jenna Marie Ortega, maybe? She played Wednesday on the Netflix-Show and she is a great actress as evidence by that.

Mary Kate Wiles aka Lydia Bennet from The Lizzie Bennett Diaries as The Doctor. She's got the right mix of humour quirkiness and underlying sadness and anger.
Giancarlo Esposito would be interesting as the Doctor OR the Master. The way he can go from lighthearted to utterly menacing would work very well for either, and although he’s been typecast as a villain, I think he could pull off a more heroic spin as well.
You know in the late 80s, Paramount wanted to do a doctor who movie with Michael Jackson? Ignoring the obvious drama that would bring in 2023, it was on the back of Thriller they thought he'd be perfect for an American movie version and tbh, I can't disagree.
Their backup plan was even more unfortunate being Bill Cosby, who is not only a horrible rapist, but would also just be a boring version of the doctor.
It might seem like an odd pick, given his physicality, but I’d go with Benjamin Byron Davis as The Doctor
He’s a big, bold performer most known for his role as Dutch van der Linde in the Red Dead Redemption duology. In that role, he was jovial and larger than life, but when the scene called for it, he could also be intimidating as all hell.
Now Davis, as The Doctor, would be unique in many ways. He’d be the first American for one, the first numbered Doctor to have facial hair, and by far the largest man to have ever held the role. Given his size, he could work action into his performance in ways no other Doctor has. I can imagine a gag with him, where he laments his Sonic Screwdriver not being able to unlock a wooden door, so he just bashes through it with his shoulder.
Benjamin Byron Davis as the Doctor, in short, would be a large, jovial man who commands attention everywhere he goes, who can also be downright scary if he needs to be.
For his companion, I have no one in particular in mind, but I just want someone who stands out from the past main companions. Maybe cast a teenager in the role, someone who, with Davis’ Doctor, could recall the grandparent-grandchild relationship from the 1st Doctor and Susan.
Casting the Master is hard. Because on one hand, Peter Blomquist as The Master is my dream. But on the other, that would mean two Red Dead actors are in the show. So, fuck it, Peter Blomquist has proven he can play evil like no other. Most famously as Micah Bell in RDR2, a slimy bastard who seems to relish in his lack of compassion. Blomquist as The Master would be a cross between the madness of John Simm and the cold, joking nature of Missy
As an American, the only American I’d want to play the Doctor is Natasha Lyonne. She’d absolutely kill it.

Edward Norton as the Master is a genius idea. But only if there’s a scene where he curb-stomps someone

His accent as Tyrion was not that great. I think you can tell he's an American by his voice. He's not played that many other British characters.
